Fouquet a resitué l'épisode du Nouveau Testament évoquant la descente du Saint-Esprit sur les fidèles assidus à la prière, dans un paysage parisien. L'assemblée est placée au niveau de l'hôtel de Nesle depuis un point de vue qui laisse découvrir la partie sud de la Cité. À droite, le pont Saint-Michel enjambe le bras de la Seine devant le Petit Châtelet. Viennent ensuite les toitures de l'Hôtel-Dieu et la tour de l'évêché, puis l'imposante façade de Notre-Dame. Devant la cathédrale s'étend le quartier dense de la rue Neuve-Notre-Dame, où se pratiquaient les métiers du livre.
